Abstract
許多研究指出出生後的幹細胞(postnatal stem cells)已成功地從許多組織中培養出來,例如骨髓、神經組織、上皮組織、骨骼肌及牙髓腔。在這些幹細胞中,牙髓幹細胞(DPSC)擁有分化成各種細胞並能發展成不同組織之能力。近年來發現到牙髓幹細胞也具有自我修復能力(self-renew),對於組織工程來說,不再是醫療廢棄物,因此可以成為很好的幹細胞來源。由於幹細胞在真正作為植入治療前,需要進行增加細胞數目,因此如何在體外加速或放大幹細胞數量一直是一個重要的研究課題。在本研究中,我們從人類牙齒中分離出牙髓幹細胞,藉由測試並觀察細胞在4000高斯之靜磁場的刺激下,對於細胞生長速度是否具有促進作用,以作為牙髓幹細胞加速增加細胞數目的方式,進一步評估靜磁場對牙髓幹細胞的貼附、細胞濃度、細胞週期、表面特徵蛋白及分化能力,並且瞭解其可能影響之訊息傳遞路徑做分析。
在實驗方法上,本研究以MTT法測試細胞的生長速度,並將培養在24孔盤的細胞分成兩組,一組放置於無充磁的磁鐵上,另一組則是放置於具表磁4000高斯之銣鐵硼磁鐵上培養96小時。我們觀察暴露在靜磁場的環境下,牙髓幹細胞的活性、細胞週期、表面特徵蛋白及分化能力並分析其訊息傳導路徑是否與MAPK pathway 相關。本研究結果顯示,牙髓幹細胞經過靜磁場的刺激後72小時增加細胞生長速度,明顯的增加了1.1倍(p< 0.05),此外,我們發現靜磁場不會影響牙髓幹細胞的型態、分化能力與表面特徵蛋白。在促進細胞生長的訊息傳導路徑方面,經由加入抑制劑的方式,本研究發現靜磁場促進牙髓幹細胞生長數目增加可能與MAPK pathway中的extracellular signal-regulated kinase (ERKs)及p38 MAPK路徑相關。
