Abstract
製造具有功能性血管網絡系統的結構一直是建立替代生物組織中的一個重大挑戰,為了克服當前組織工程的困難,我利用三維生物打印策略來開發生物墨水。當中,生物墨水材料由載有生長因子的光交聯甲基丙烯酸明膠(Gelatin Methacrylate, GelMA)複合水凝膠組成,並於血液來源的人臍靜脈內皮細胞(Human umbilical vein endothelial cells, HUVECs)和脂肪來源幹細胞(Adipose-derived stem cells,ADSCs)共同培養之水凝膠模型中形成穩定的脈管系統網絡,研究了從內皮細胞形成血管網絡的新興組織工程方法。本研究提出GelMA與血管生長因子相結合,之後與HUVECs以及ADSCs進行細胞包覆(Cell encapsulation),以模擬體內血管生成和發育過程,並誘導血管組織新生。對於GelMA、GelMA@VEGF、GelMA@FGF這三種不同水凝膠分別以Cell Counting Kit 8 Assay Kit (CCK-8)進行活性測試,在細胞包覆後培養第一周內,微血管網絡形成並保持功能穩定,此後,細胞逐漸形成管腔,在第二週後分化為血管周圍細胞以穩定血管,整個構建體在體外培養期間皆表現出高結構穩定性,驗證細胞在具有生長因子結合之材料中能幫助細胞增長。此外,亦以螢光染色標誌去辨識內含DNA的細胞核及粒線體的位置,由結果證明生長因子水凝膠促進 HUVECs及ADSCs血管生成的潛力,得出此建構體能夠有效支持細胞存活和增殖。
