Abstract
過量精製糖造成負面健康影響使非營養性甜味劑的使用量逐漸增加。過去文獻指出人工甜味劑可能增加體內氧化壓力,對於使用非營養性甜味劑為主的女性可能造成不利影響。因此,天然萃取甜味劑 (如:甜菊糖苷、羅漢果苷) 受到重視,然而其對於女性生殖系統之影響及作用仍不清楚。本次研究探討甜菊糖苷、羅漢果苷之代謝物對於顆粒細胞之影響。實驗以人類顆粒樣腫瘤細胞 (KGN) 介入1、5、10 µM之甜菊醇酰基葡萄糖醛酸及羅漢果醇24小時,以螢光染色檢測細胞及粒線體氧化壓力、粒線體數量及膜電位,以比色法分析抗氧化酵素及氧化壓力指標,以生物能量測定儀評估粒線體功能,以聚合酶鏈反應評估類固醇賀爾蒙生合成相關mRNA表現,以西方點墨法測量粒線體生合成相關蛋白質表現量以及以免疫呈色法評估黃體素分泌量。結果說明,甜菊醇酰基葡萄糖醛酸並未顯著改變Bucladesine (dB-cAMP) 誘導的黃體素生合成,但顯著增加粒線體氧化壓力及 Catalase (CAT) 活性,降低粒線體儲備呼吸率。羅漢果醇顯著降低 dB-cAMP誘導的黃體素分泌,其可能透過磷酸化AMP-activated protein kinase (AMPK) 後降低Extracellular signal-regulated kinase 1/2 (ERK1/2) 磷酸化進而減少3-beta-hydroxysteroid dehydrogenase (3β-HSD) 之mRNA表現量,同時也增加細胞及粒線體中氧化壓力,代償性上調粒線體生合成。
