Abstract
酒精性肝臟疾病 (alcoholic liver disease, ALD) 在世界各地都是主要的健康問題,而研究發現牛樟芝 (Antrodia camphorata) 能夠延緩酒精性肝損傷,其中的活性成份 antroquinonol (ANQ) 具抗發炎、抗氧化特性,然而對於 ALD 是否具改善作用尚不明瞭,因此本研究利用酒精誘發肝臟損傷之動物模式以探討 ANQ 對 ALD 的影響並探討其中可能之影響機制。首先為釐清 ANQ 對 ALD 的影響,利用灌食酒精的方式探討 ANQ 是否會直接影響酒精的吸收。接著為探討 ANQ 在 ALD 中是否具有保護肝臟的作用,利用短期酒精攝取之動物模式觀察 ANQ 對於酒精損傷的影響。由於我們發現 ANQ 確實可能有助於改善 ALD 的進程,且其作用並非來自直接影響酒精的吸收,因此我們進一步以長期慢性酒精攝取之動物模式探討飲食中 ANQ 的補充改善 ALD 之可能機制。實驗結果顯示補充 ANQ 有助於改善肝臟損傷,並可能藉由降低 CYP2E1 與提升 Nrf-2 表現以達到抗氧化作用、透過抑制 TLR4 訊號傳遞路徑以抑制發炎反應、經由調控 AMPK 及其下游蛋白質表現以改善肝臟脂質代謝。綜合以上結果顯示 ANQ 有助於肝臟氧化壓力、發炎反應與脂質代謝的改善因而可能達到延緩 ALD 疾病進程的作用。
