Abstract
高血壓為全球重視之公共健康議題,由於患者平時感受不到明顯症狀而有隱形殺手之稱,高血壓為心血管疾病、腦血管疾病及腎臟疾病的重要危險因子,而內皮功能、氧化壓力及發炎反應在高血壓疾病中扮演重要的角色,若發生病變及異常皆會導致高血壓的情形加劇並導致罹患併發症的風險及死亡率增加。Antroquinonol 為牛樟芝菌絲體中的泛醌類衍生物,具有抗癌、抗氧化及抗發炎的作用,因此本實驗利用 L-NAME 誘發內皮功能缺損之高血壓動物模式探討 antroquinonol 的介入是否有助於改善高血壓情形並進行機制探討。實驗使用 40 隻雄性 Wistar 大鼠,進行為期 9 週的實驗,分為以下 4 組:控制組、L-NAME 組 (於飲水中添加每公斤體重 25 毫克 L-NAME)、低劑量及高劑量 antroquinonol 組 (飲水為 L-NAME 飲水,並於飲食中添加每公斤體重 20 及 40 毫克 antroquinonol)。實驗結果發現,L-NAME 會快速的使血壓上升,並使體內氧化壓力及發炎反應情形加劇;Antroquinonol 的介入則可以有效延緩血壓的上升,並透過調節體內血管活性因子降低腎臟 NOX 之活化並刺激 Nrf2 之活化以增加抗氧化物質的產生,另外透過抑制 NFκB 的活化以減少促發炎細胞激素產生。本實驗結果顯示 antroquinonol 的介入有助於降低實驗動物腎臟之氧化壓力及發炎反應,而可能達到改善血壓及內皮功能與腎臟保護的作用。
