Abstract
結直腸癌是世界上最普遍的惡性腫瘤,也是癌症相關死亡的第二大常見原因。 5-氟尿嘧啶等細胞毒性化療藥物是臨床治療癌症的關鍵藥物,從1960年代開始單獨使用5-氟尿嘧啶治療,到現今許多藥物組合仍是以5-氟尿嘧啶作為基底,包括:FOLFOX (活性葉酸、5-氟尿嘧啶、雙胺環己烷草酸鉑)、FOLFIRI (活性葉酸、5-氟尿嘧啶、愛萊諾迪肯) ,不僅提高了反應率,也延長了整體存活期,但研究發現結直腸癌的預後仍然較差。 可能是耐藥性的頻繁發生限制了抗癌治療的療效,因此鑑定耐藥性對於尋找結直腸癌的治療方法具有重要意義。在這項研究中,我們從 Gene Expression Omnibus (GEO) 數據集 (GSE77180) 和 具抗藥性細胞的次世代定序 (NGS) 中鑑定了5-FU抗藥性基因的差異表達。 在本研究的實驗方法中,我們首先建立了長期的抗5-FU結直腸癌細胞,並探索了結直腸細胞於5-FU治療中的改變。將對來自數據庫比較的重疊基因進行抑制或過表達,以觀察基因在細胞中的表達。 結果表明,抑制5-FU抗藥基因(APOBEC3B、APOBEC3H、CCNE2和COL12A1)可降低結直腸癌細胞HCT116的生長,反之,外送5-FU抗藥基因可增強細胞生長和對5-FU的耐藥性。總結來說,本研究結果表明APOBEC3B、APOBEC3H、CCNE2和COL12A1在結直腸癌細胞的5-FU抗藥性細胞中扮演至關重要的作用,這將是克服結直腸癌抗藥性有前途的治療策略。
