Abstract
本研究開發可遞送粒細胞-巨噬細胞集落刺激因子(GM-CSF)的可注射型光感性明膠水膠,作為吸引樹突細胞的載體使應用於癌症免疫治療。首先,利用甲基丙烯酸酐 ( methacrylic anhydride, MA )與明膠合成甲基丙烯酸酯化明膠 ( Gelatin methacryloyl, GelMA ),然後將此溶液經光引發自由基聚合形成共價交聯的甲基丙烯酸酯化明膠水膠 ( GelMA hydrogel )。
GelMA以不同濃度 ( 5,7.5,10,15和20 w / v % )配置形成GelMA 水膠載體,以流變學試驗和降解試驗評估其物化性質。結果顯示,當GelMA濃度提高時,可以增加水膠的穩定性和機械性能並延長水膠降解時間從5天內延長至30天以上。在動物實驗結果也證實當水膠注射至老鼠背部皮下後,水膠會隨濃度增加導致降解減緩。
經由物理性質,體外和體內實驗結果顯示水膠最佳化處方為GAD0.1C15I8.8 和GAD0.1C20I14,其水膠處方為使用type A種類的明膠,以MA和明膠為0.1/1的比例合成,並分別配製成15和20 %濃度然後再以8.8和14 mW/cm2之照光強度形成。將上述兩個最佳處方進一步包覆GM-CSF經光引發自由基聚合形成裝載GM-CSF之明膠水膠 ( GelMA/GM-CSF hydrogel )。評估這個水膠體外的藥物釋放,結果顯示GAD0.1C20I14水膠藥物釋放在第7天才達到100 %,所以相較於GAD0.1C15I8.8水膠,GAD0.1C20I14水膠為較佳之處方。在體外樹突細胞遷移試驗以GAD0.1C20I14/GM-CSF 水膠評估其吸引樹突細胞能力,結果證實通過釋放GM-CSF可成功吸引6.7 %的樹突細胞至水膠中。
所以這種裝載GM-CSF的可注射的光感性水膠可通過局部調控樹突細胞,增強其抗原攝取和呈現的機會,因此對於癌症免疫治療展現出很大的潛力。
