Monocyte chemotactic protein-induced protein 1 (MCPIP1) 是一種新穎CCCH 型鋅指蛋白質,在巨噬細胞的發炎反應中扮演必要的調控者的角色。在本篇的研究中,藉由誘導系統使MCPIP1 過度表現在人類腎臟胚胎 T-REx-293 細胞中,我們發現MCPIP1能促進 tumor necrosis factor α (TNFα) 所誘導的死亡。利用流式細胞儀分析,得知 MCPIP1 可增強 TNFα 所引發的sub-G1 波峰;然而,突變型 MCPIP1 在 TNFα 的處理下並沒有增強sub-G1 波峰。我們觀察到野生型 MCPIP1 而非突變型 MCPIP1 能增強 TNFα 處理細胞後,對 caspase-3, -6, -7, -8, -9 的活化和加強 Poly (ADP-ribose) polymerase (PARP) 的切割。更發現野生型 MCPIP1 能促進 TNFα 對細胞凋亡前因子, c-Jun NH2-terminal kinase (JNK) 磷酸化表現的增加。有趣的是,野生型 MCPIP1 在 TNFα 處理後會被分解,此一情形會因 capase-1 抑制劑的存在而被回復。此外,我們發現 MCPIP1 具有被 capase-1 切割的保留性序列。總而言之,我們的結果顯示 MCPIP1 能透過活化 caspases cascade,增強 TNFα 所誘導的細胞凋亡路徑訊息傳遞路徑。
