Abstract
冬蟲夏草(Cordyceps sinensis,CS)為在亞洲地區廣泛使用於傳統
中醫及健康食品。目前已有許多冬蟲夏草內的活性成分,例如:
Cordycepin,固醇類以及多醣體萃取物已被證實可影響發炎狀況、神
經系統、呼吸以及心血管疾病等。其中多醣體為主要抗氧化成分,並
且已被證實具有抗發炎、抗腫瘤以及免疫調節的功能。CME-1,係一
由冬蟲夏草菌絲體所萃取出的多醣萃取物,其主要組成為半乳糖和甘
露糖,分子量為27.6 kDa。目前已被證實具有抑制過氧化氫所誘發之
細胞死亡以及抗自由基形成。
血栓在許多心血管疾病,例如:中風以及心肌梗塞中扮演著很重
要的角色,而血栓的形成主要與血小板的活化有關,因此,目前臨床
上許多抗血小板藥物已使用於心血管疾病的治療。然而CME-1 目前
對血小板的作用尚未明確,因此我們有意探討CME-1 在血小板活化
過程中所扮演的角色。
先前的實驗已證實,CME-1 隨著濃度的增加(125-210 ?慊/ml)能有
效抑制collagen 所引起的血小板活化,此現象並非透過與collagen 競
爭GP IIb/IIIa receptor,其機轉可能是經由抑制collagen 下游路徑,包
含了抑制血小板內PLC??2、p47 protein、Akt 以及mitogen-activated
protein kinases (MAPKs)的磷酸化。延續先前的研究,本次實驗發現,

CME-1 (125-420 ?慊/ml)除了不會抑制collagen receptor (GP VI 和
integrin ??2??1)外,CME-1(125 和210 ?慊/ml)也可增加血小板內部cyclic
AMP 的形成,但不會影響cyclic GMP,達到抑制血小板凝集,亦會
引起vasodilator-stimulated phosphoprotein (VASP)的磷酸化,此外
CME-1 (62.5-210 ?慊/ml)還可有效抑制低濃度刺激劑(eg. thrombin、AA)
所誘導的血小板凝集以及血小板內部的訊息傳遞路徑,包含了
MAPKs 以及Akt 的磷酸化。而當CME-1 與SQ22536 (adenylyl cyclase
抑制劑)合用時,p38 MAPK 以及Akt 的磷酸化有被回復的現象。
最後,由我們的實驗結果首次發現,CME-1 是透過增加血小板內
部的cyclic AMP 濃度,進而抑制血小板內部p38 MAPK 以及Akt 的
磷酸化來達到抗血小板的作用。因此CME-1 也許具有治療心血管疾
病的潛力。
