Abstract
中文摘要
Hispolon(HIS)是天然菌菇桑黃(PL)的主要成分,已有文獻指出桑黃(Phellinus linteus)會抑制腫瘤生長和抗氧化作用,但是目前對於Hispolon在NO以及iNOS之抗發炎機轉方面的研究仍不清楚。因此,在本研究中,以巨噬細胞RAW264.7細胞為實驗模式,首先利用hispolon誘導HO-1蛋白表現而抑制酯多醣(Lipopolysaccharide, LPS), 壁脂酸(Lipoteichoic Acid, LTA),及聚醣(Peptidoglycan,PGN) 格蘭氏陰性菌及格蘭氏陽性菌細胞壁上之內毒素。這些細菌內毒素誘導NO生成。而利用ROS抑制劑NAC發現Hispolon透過ROS誘導HO-1的mRNA以及蛋白的表現。此外,利用HO-1抑制劑 SnPP及HO-1 siRNA與Hispolon及內毒素共同處理後,Hispolon抑制LPS或LTA及PGN誘導NO生成明顯減弱。Hispolon透過調節轉錄因子NFkB及AP-1活性而抑制iNOS/NO表現,且抑制內毒素所活化的磷酸化JNK蛋白表現而在研究細胞凋亡方面,利用 MTT 試驗發現 Hispolon 能夠有效的抑制由格蘭氏陰性菌及格蘭氏陽性菌細胞壁上之內毒素所誘導的細胞凋亡。然後進一步利用 DNA 片斷化電泳分析證實了 Hispolon 的確能夠抑制由LPS或LTA及PGN誘導NO走向凋亡而減少其存活率。
綜合以上的實驗結果,本篇論文認為 Hispolon 能夠透過抑制一些細菌內毒素如LPS或LTA及PGN而保護細胞達到抗發炎以及抗細胞凋亡的功效,因此,Hispolon 應具有可開發成為抗發炎新生藥物的潛力,期望未來能夠將其真正開發為藥物,為對抗發炎疾病貢獻一份力量。
