Abstract
行政院衛生福利部2014年四月所公布之台灣2012年最新國人十大癌症,女性罹患卵巢、輸卵管及寬韌帶之癌症為女性罹癌率的第九名,而其致死率則為第八名。在卵巢癌轉移前及早給予病患治療,其五年平均存活率為92%。雖然早期介入對於治療卵巢癌病患具有顯著療效,但卵巢癌之抗藥性已於臨床出現,因此,發展新的治療藥物是非常重要的。已有研究證實蒽醌可作為不同的臨床應用,包括:通便、抗癌、抗發炎、抑菌、利尿、血管舒張⋯⋯等。因此本研究旨在探討新型蒽醌類化合物用於具抗藥性卵巢癌細胞的機制。
利用MTT 測試比較各種癌細胞在藥物處理後之存活率,結果顯示A2780R2000對三種藥物均具有感受性,而compound I、II、III的IC50分別為33.44μM、25.77μM、24.59μM。接著,利用細胞週期分析(Cell cycle analysis)分析在藥物處理後的不同時間的細胞週期變化,發現藥物使細胞在早期時其細胞週期會受到抑制,而於晚期時誘導細胞走向細胞凋亡(Apoptosis)。最後,經由西方墨點法之結果發現藥物可能是藉由DNA的破壞導致細胞週期的停滯。因此,研究結果證實新型蒽醌類化合物對於臨床化學治療是一個具潛力之抗癌藥物。
