Abstract
關節軟骨(Articular cartilage)屬於一種難以自行修復的組織,且近年來關節軟骨損傷患者的年齡層有逐漸下降的趨勢。且隨著生醫科技的持續進展,模擬關節軟骨微環境的水凝膠(Hydrogel)研究也日益受到關注,主要歸因於其高度含水量及良好的可客製化物理特性。
因此,本研究以合成高分子聚乙二醇(Polyethylene Glycol, PEG)與聚乙二醇二丙烯酸酯(Poly(ethylene glycol) diacrylate, PEGDA)依不同比例形成水凝膠基質,利用其高生物相容性與低細胞毒性,分別添加羥基磷灰石(Hydroxyapatite)粉體與含1%慶大黴素之羥基磷灰石粉體,製備具藥性之水凝膠材料,探討其材料特性與生物表現。
在初期實驗階段,比較不同比例PEG/PEGDA配方後發現,隨著 PEG含量增加及 PEGDA含量減少,水凝膠的溶脹率顯著提升,然而,在抗壓測試中亦觀察到PEG含量增加會導致材料機械強度下降。為改善此現象,後續實驗中加入大豆磷脂酰膽鹼(L-α-Phosphatidylcholine)對粉體進行改質,以提升其在水中之分散性,進而增強水凝膠整體之機械強度;同時亦將藥性粉體中慶大黴素含量由 1% 提高至 5%,以觀察藥物釋放之效果。
後期實驗結果顯示,增加藥物含量確實可促進慶大黴素從水凝膠中的釋放,且改質後粉體亦顯現較佳的機械強度。然而,於含 5% 慶大黴素的樣品中,卻未觀察到藥物釋放現象,顯示仍需進一步探討其影響機制。
綜合上述結果可知,採用聚乙二醇為基質並搭配光固化技術所製備之水凝膠,於材料設計與藥物釋放控制方面皆展現出極具潛力之應用前景,值得進一步優化與研究。
