Abstract
大腸癌為台灣癌症發生率的第一名,流行病學指出攝取全穀類可降低罹患大腸癌的風險,其中小米 (Setaria italica) 亦為富含多酚類且抗氧化能力高之全榖類,有助於大腸癌的預防。已有細胞研究指出小米多酚萃取物介入下,能有效預防大腸癌細胞生長,但目前在小米介入對大腸癌的動物實驗相關研究仍有限,因此本次擬以1,2-dimethylhydrazine (DMH) 誘發大鼠大腸癌之動物模式,探討小米介入對大腸癌前期病變生成之影響。實驗將雄性Fisher 344大鼠分為6組:其中Normal control (N) 組與DMH (D) 組餵食AIN-93G標準飼料,LT組以10%台東8號小米 (Taitung No.8 millet) 取代標準飼料,HT組以30%台東8號小米取代標準飼料,LK組以10%基努李西亞小米 (Keanu Lishia millet) 取代標準飼料,HK組以30%基努李西亞小米取代標準飼料。除N組外,於實驗期第一週起,每週以腹腔注射方式給予40 mg/kg大腸癌致癌物DMH,共注射4劑。經過10週的實驗期後,發現低、高劑量台東8號小米與基努李西亞小米皆顯著減少異常腺窩病灶 (aberrant crypt foci, ACF)、sialomucin-producing aberrant crypt foci (SIM-ACF) 及黏液素缺乏病灶 (mucin-depleted foci, MDF) 數,另顯著提升大腸組織抗氧化相關酵素superoxide dismutase (SOD)、glutathione peroxidase (GPx) 及catalase (CAT) 之活性及促細胞凋亡相關蛋白Bcl-2-associated X protein (Bax) 表現,並降低抑細胞凋亡相關蛋白 B-cell lymphoma/leukemia 2 (Bcl-2)、細胞增生指標proliferating cell nuclear antigen (PCNA) 之表現,以上結果顯示小米具預防大腸癌前期病變之效果,其作用機制可能與調控抗氧化能力及調控凋亡相關蛋白表現有關。
