Abstract
微生物轉換為利用微生物的整體細胞或分離的酵素對受質進行結構修飾和改變以得到新的有機化合物。Diosgenin (1)具spirostan骨架,結構與類固醇類似,為製藥工業合成性荷爾蒙之原料,此外並具有多種生物活性。由於diosgenin (1)具特異的多環骨架,適合以微生物轉換來修飾其構造,因此,為了開發具新的藥理活性之化合物,先將diosgenin (1)化學修飾為2,接著進行微生物轉換研究。經由15株菌種篩選,選擇Bacillus megaterium及Streptomyces griseus 進行大量發酵培養。受質2與B. megaterium培養得到化合物3?{9;受質2與S. griseus培養得到化合物3、10?{12,其中化合物4、6?{8與10?{12為新的化合物。所得化合物皆經由一維、二維核磁共振光譜及高解析質譜鑑定其結構;分離得到的化合物將進一步進行生物活性試驗探討。
