Abstract
骨骼肌損傷與退化 (muscle injury and atrophy) 是老化、神經損傷及疾病相關治療(如化療藥物)常見的併發症。粒線體在調控細胞能量代謝、抗氧化壓力 (Anti-oxidative stress) 及細胞存活中扮演關鍵角色。本研究探討粒線體移植 (mitochondrial transplantation, MT) 作為治療細胞毒性誘導之骨骼肌損傷 (cytotoxic-induced skeletal muscle injury) 的潛在策略及其作用機制。在細胞毒性環境下,粒線體功能障礙導致 ATP 生成減少、氧化壓力增加及肌肉細胞生長及代謝能力下降。因此,本研究假設粒線體移植能夠恢復受損肌肉細胞的能量代謝,改善細胞存活並促進肌肉細胞分化及增生。本研究首先針對粒線體的分離與保存條件進行確認,並透過 pDsRed2-Mito 標記追蹤粒線體,也針對分離出的粒線體進行功能性測試。其次,建立細胞毒性模型透過 Oligomycin 誘導粒線體損傷,並評估粒線體移植對 C2C12 骨骼肌細胞之影響,包含細胞生長、代謝狀況、細胞內氧化壓力以及相關基因表達,以了解肌肉修復與粒線體之間的關聯性。實驗結果顯示,粒線體移植能提高受損細胞的存活率與 ATP 生成,並促進細胞增殖,螢光標定證實移植的粒線體成功與宿主細胞整合。此外,粒線體移植可能透過調控氧化壓力,以及調節骨骼肌再生相關基因 (MyoD、MyoG、Cav3) 來促進肌肉功能恢復。本研究證明粒線體移植有望做為治療細胞毒性誘導之骨骼肌損傷的策略,未來可進一步探討粒線體移植於因神經性損傷造成肌肉退化的治療潛力。
