Abstract
2019 年爆發的嚴重特殊傳染性肺炎(COVID-19)對人類產生了巨大的影響。為 了對抗引起這次疫情的嚴重急性呼吸道症候群冠狀病毒 2 型(SARS-CoV-2),mRNA 平台取得了重大突破,對疫苗開發做出了重要貢獻,而已發表的研究證明了疫苗的有 效性和安全性。雖然適應性免疫受體組庫能反映接種疫苗後的記憶反應,卻只有少數 研究從適應性免疫受體組庫的角度討論了疫苗對人體的影響。目前已發表的免疫組庫 相關研究中,皆著重討論「接種疫苗前後」,關於「加強劑量」的重要性,並未有研 究針對這方面進行討論。為了了解疫苗加強劑量接種後的反應,我們的研究從 29 名健 受試者中構建了 T 細胞受體(TCR)組庫,進行了施打第二劑 Moderna COVID-19 疫苗 (Spikevax,mRNA-1273)前後的組庫變化比較。我們使用了 5'RACE 搭配 oligo 以及 nested TCR 進行建庫、次世代定序(NGS)進行序列測定、並使用 R 語言進行數據分 析。本研究的目的是比較 29 名受試者接種 Moderna COVID-19 疫苗的第二劑前後的 T 細胞受體特徵;比較與 SARS-CoV-2 相關的 T 細胞受體與整體 T 細胞受體特徵的差異 了解第二劑 Moderna COVID-19 疫苗接種後擴增 T 細胞受體的共同特徵;並了解施打疫 苗後的反應原性和 T 細胞受體特徵之間的關係。結果顯示,從許多角度探討第二劑 mRNA-1273 施打前後的 T 細胞受體並沒有顯著差異;相較於整體 T 細胞,SARS-CoV-2 相關的 T 細胞受體,CDR3 顯著較短、並且有 6 個 TRBV 和 4 個 TRBJ 基因有顯著較高 的使用;在所有受試者當中有觀察到相似特性的 T 細胞受體,且相較於非擴增 T 細胞, 可以辨識到 S240-282 和 S684-726 的 T 細胞受體在擴增 T 細胞的出現率顯著較高;此 外,在施打疫苗後,發生注射部位紅疹和肌肉痛的受試者中觀察到更高的 TRB 多樣性。 整體而言,儘管第二劑接種在 T 細胞受體特徵的角度上似乎沒有顯著效果,但在特定 表位的 T 細胞反應中仍然存在顯著增加,並且在所有受試者中仍然觀察到第二劑接種 後的共享 T 細胞受體擴增。第二劑接種的有效性在 T 細胞受體特徵的角度上仍然存在, 儘管不是顯著的。這篇研究的圖示摘要呈現於 Figure 1。
