Abstract
根據統計女性乳癌一直是10大癌症中的首位,而且在比例上還有不斷攀升的趨勢,其中又以三陰性乳癌最為難治,惡性度高不易治療,三陰性型乳癌顧名思義就是沒有 ER、PR和 HER2 受體,所以這類型的癌症對於病患無法使用賀爾蒙療法治療,缺乏 HER2受體也使的無法使用針對HER2的標靶藥物。晚期三陰性乳癌患者存活期通常不到一年半,更糟的是,復發後死亡率很高,以林口長庚2萬筆統計資料顯示,三陰性乳癌患者一旦病情復發,平均5年內存活率12% 。目前臨床上沒有針對有效之療法,本實驗透過高轉移活體動物乳癌DNA微陣列資料庫中與自噬作用中不同階段,啟動成核(Nucleation)、自噬小體延伸(Elongation)及降解(Degradation)相關基因DNA表現,分析出高轉移性三陰性乳癌細胞2nd lung met MDA-MB-231在SQSTM1, BECN1, PI3KC3相較於MDA-MB-231表現量較高且具有顯著差異,並且以基因剔除技術CRISPR/Cas9為基礎建立2nd lung met MDA-MB-231 ATGs剔除模型, 並且利用飢餓模式作為引發自噬作用的工具。並藉由LC3-II的蛋白表現與LC3 puncta作為確認引發自噬作用的方法,最後在ATGs基因剔除的2nd lung met MDA-MB-231中引發自噬作用。經過分析細胞凋亡標誌、細胞週期抑制標誌發現在自噬作用發生情況下但ATGs無法作用時,會造成細胞無法進行細胞凋亡反應。反過來思考,當癌細胞要正常進行凋亡需要借助ATGs的作用,因此也許在使用凋亡性藥物在治療三陰性乳癌時可以同時施以自噬作用引發劑加強癌細胞之凋亡反應。
