Abstract
慢性骨髓性白血病 (Chronic Myeloid Leukemia, CML) 屬於骨髓增生性疾病之一,由費BCR-ABL致癌蛋白所造成。BCR-ABL是個持續活化態tyrosine kinase,BCR-ABL會誘導細胞增生、抑制凋亡及阻止分化。本實驗室之前已經利用microarray篩選出雙特異性磷酸酶21 (Dual specificity phosphatases 21, DUSP21) 是一個imatinib inducible gene。過去研究顯示,DUSP21屬於DUSPs家族的一員,於肝癌細胞中,扮演細胞增生的角色,但DUSP21在CML中扮演的角色是未知的。因此在本論文探討於CML細胞中此基因的功能角色。RT-PCR實驗證實於K562 CML細胞DUSP21的表現隨著imatinib濃度和時間的增加而增加。此外imatinib誘導KU812 CML細胞的紅血球分化基因和DUSP21的表現。在對imatinib有抗藥性的BaF3/T315I細胞,imatinib並不會誘導DUSP21表現,但給予另一個BCR-ABL inhibitor,ponatinib,發現會使K562、BaF3/p210以及BaF3/T315I細胞的DUSP21 表現增加。Ponatinib並且會誘導K562細胞紅血球分化基因的表現。Imatinib和ponatinib皆會誘導CML細胞株的DUSP21蛋白質表現。Activin A (紅血球分化因子)會誘導DUSP21以及其他紅血球分化基因,glycophorin A、alpha globin、zeta globin、erythroid specific transcription factors: GATA1和NF-E2p45的表現。為了證明DUSP21是否參與imatinib誘導的紅血球分化,將K562細胞的DUSP21 knockdown後再處理imatinib,RT-PCR的結果顯示紅血球分化基因的表現下降;反之,DUSP21 overexpression,紅血球分化基因表現會增加。DUSP21 knockdown,GATA1 promoter活性會下降;另外當DUSP21大量表現,GATA1 promoter活性會增加。進一步實驗證明,不管BaF3/p210或BaF3/T315I細胞大量表現DUSP21時,會對ponatinib的敏感性增加。
本研究結果顯示,在慢性骨髓性白血病細胞中,imatinib和ponatinib會誘導DUSP21和紅血球分化。DUSP21大量表現會增加紅血球分化基因表現以及使imatinib敏感性和抗藥性的CML細胞對ponatinib產生敏感性。
