Abstract
很多來自植物及草藥的重要化合物具有抗氧化活性,但其抗氧化作用機制各有不同。咖啡酸相關衍生物及冬蟲夏草菌絲體的多醣體萃取物的生物活性被探討很多,並專注在其抗氧化能力。本論文探討天然化合物抗氧化的分子機制,期待藉著對分子作用的了解能進一步改善對細胞或活體的生化調控。CME-1係一新穎冬蟲夏草菌絲體多醣體萃取物,首度的被分離與鑑定,其係一主要以半乳糖與甘露糖組成、分子量為27.6 KDa的低分子量多醣萃取物。CME-1呈現對過氧化氫處理之巨噬細胞的保護作用,同時也觀察到其降低細胞內氧化壓力與維持粒線體膜電位之活性。為了分析此一保護機制,分別探討其清除過氧化氫、與抑制生成活性氧酵素活性之功效與能力。結果顯示CME-1保護巨噬細胞的作用與其過氧化氫之清除功效並無顯著相關性。進一步探討其對各種生成活性氧酵素活性之調節能力,結果顯示其對黃嘌呤氧化酶未具顯著的抑制能力;相對的,對於中性及酸性鞘磷脂酶活性則具有顯著的抑制活性。隨著過氧化氫處理,巨噬細胞內的鞘磷脂酶產物—神經醯胺隨之大量增加,CME-1共同處理時顯現神經醯胺量的明顯降低。推論透過對細胞內神經醯胺訊息傳導途徑的阻礙作用是CME-1的重要功能,因此揭露天然多醣類化合物調節巨噬細胞的分子目標,此將提供在此一領域的研發工作者於評估多醣類化合物功效的依據。為了結合不同抗氧化機制以擴大有效的範圍,本論文也探討不同親水性性質的咖啡酸衍生物作用於黃嘌呤氧化酶的抑制機制,結果顯示不同親水性性質官能機的衍生物結合在黃嘌呤氧化酶的不同功能區,同樣達到抑制黃嘌呤氧化酶活性之功效。綜此,咖啡酸衍生物具有清除活性氧及黃嘌呤氧化酶抑制活性;而CME-1則可抑制鞘磷脂酶活性、阻礙神經醯胺訊息傳導途徑,結合不同的抗氧化機制的組合處理,將提供醫藥應用的新機能。
