Abstract
Stevioside為甜菊[Stevia rebaudiana (Bertoni) Bertoni]葉中主要的diterpenoid glycoside,可作為甜味劑,且具多種藥理活性。Steviol (1)及其類似物3具有ent-kaurene之骨架,皆衍生自stevioside。文獻指出,Steviol (1) 具抗增殖、免疫調節及作用於?? cells以刺激胰島素分泌等活性。由於此兩化合物具有isoprenoid支鏈骨架,適合利用酵素來修飾其結構,因此為了獲得更多ent-kaurene之化合物以開發新的生物活性,選擇Steviol (1)與3為受質進行微生物轉換研究。經由菌種篩選及再現性試驗結果,選擇Mortierella isabellina、Beauveria sulfurescens與Nocardia sp.進行微生物轉換。研究發現受質steviol (1)經由與M. isabellina培養得到代謝物ent-13,16??,17-trihydroxy-kauran-19-oic acid (4)、5、ent-7??,13-dihydroxy-kaur-16-en-19-oic acid (6)、7與ent-9??,13-dihydroxy-kaur-16-en-19-oic acid (8);另經由與B. sulfurescens培養得到4、ent-13,17-dihydroxy-kaur-15-en-19-oic acid (9)與10。另一方面,受質3 經由與Nocardia sp.培養得到ent-13-hydroxy-16??-methyl-15-oxokauran-19-oic acid (11)、ent-13-hydroxy-16??-methyl-15-oxokauran-19-oic acid methyl ester (12)、13和14;另經由與M. isabellina培養得到2、11與15。其中,代謝物5、7、10、13與14為新化合物。所得之新化合物皆經由一維、二維核磁共振光譜及高解析質譜儀鑑定其結構。此外,分離所得之化合物將進行藥理活性試驗。
