Abstract
國人飲食西方化,長期攝入過多精緻糖類、脂質及膽固醇時,使得體內脂肪、膽固醇堆積與活性氧物質增加,促使粥狀動脈硬化發生,進而導致心血管疾病。近年來有研究指出,心血管疾病之進展與粒線體功能及損傷有關。薑黃素為一多酚類化合物,具有抗氧化、抗發炎、促進癌細胞凋亡及抑制腫瘤增生等益處。本次研究目的為探討西方飲食補充薑黃素對於心血管疾病危險因子、抗氧化酵素、粒線體功能及內皮性一氧化氮生合成作用之影響。實驗將六週齡雄性 SD 大鼠隨機分為兩組 : 控制組 (C) 與西方飲食組 (W),進行八週飲食介入。八週後,再將控制組隨機分為兩組 : 正常飲食組 (C) 與正常飲食再添加薑黃素組 (CC),而將西方飲食組分為四組 : 西方飲食組 (W)、西方飲食再添加薑黃素組 (WC)、改為正常飲食組 (WN) 與改為正常飲食再介入薑黃素組 (WNC),介入四週薑黃素 (300 mg/kg BW/day) 後予以犧牲,取其血液、心臟、主動脈進行分析。實驗十二週後,WC組相較於W組能顯著提升一氧化氮、降低黏附因子sI-CAM-1與sV-CAM-1、Hcy、MDA含量,並顯著提升抗氧化酵素活性、粒線體電子傳遞鏈酵素複合體活性與粒線體功能指標,同時亦顯著增加心臟、血管中內皮性一氧化氮合成酶蛋白質表現。因此,薑黃素可降低西方飲食所誘發之氧化傷害,提升抗氧化酵素活性,減少脂質過氧化,進而改善粒線體功能受損,並增加內皮性一氧化氮生合成作用與降低粥狀動脈硬化指標,以達到改善心血管疾病危險因子及保護心血管之作用。
