Abstract
細胞色素 P450 17A1(CYP17A1)在雄性激素生成途徑中將孕烯醇酮(pregnenolone)或黃體素(progesterone)的 D 環上 17 號碳原子上 α 位加上一個羥基,生成 17α-羥孕烯醇酮(17α-hydroxypregnenolone)或 17α-羥孕酮(17α-hydroxyprogesterone),之後斷裂 17 號碳原子與 20 號碳原子之間的碳碳鍵,生成去氫表雄酮(dehydroepiandrosterone,DHEA)或雄烯二酮(androstenedione),對於男性體內雄性激素的生合成扮演重要的角色。阿比特龍(abiraterone)為細胞色素 P450 17A1 抑制劑,通過美國 FDA 之核可用於藥物或手術去勢抗性的轉移性攝護腺癌,但長期使用仍有藥物耐受性的問題,因此若在雄性激素的控制上發展出雙藥效藥物,將有助於轉移性攝護腺癌有效控制。本研究以阿比特龍為先導化合物進行結構修飾。先導化合物在P450 17A1 抑制活性為 μM 等級,卻意外發現在對抗攝護腺癌細胞株細胞毒性測試上達到 nM 的等級,本研究發展具有雙效癌症抑制之機轉化合物,推測有潛力成為對抗攝護腺癌之候選藥物。
