Abstract
在所有的癌症當中,肺癌的死亡率高達15-20%,儘管醫學進步,肺癌的存活率在過去十年並沒有大的改變,不論男女,在發病率和死 亡率中,肺癌排名都是第一位,其中癌轉移是造成治療失敗一個很重要的因素,由於造成癌轉移的機轉尚未明瞭,故本實驗室利用兩株高轉移力肺腺癌細胞(CL1-5F4)與低轉移力肺腺癌細胞(CL1-0)希望可以尋找到與肺腺癌轉移相關的基因,可以在癌細胞轉移前及早發現及早治療,增加肺腺癌的存活率。
利用Subtraction hybridization的方法,比較淋巴結轉移之肺腺癌與原位癌的病人組織,找到14個與轉移相關的基因,以CL1-0和CL1-5F4細胞株當做細胞模型,用反轉錄聚合酵素連鎖反應【reverse transcription polymerase chain reaction(RT-PCR)】以及即時定量反轉錄聚合酵素連鎖反應(Real-Time reverse transcription PCR)比較上述14個基因表現量的差異,我們發現Regulator of G Protein signaling protein(RGS11)mRNA的表現量在CL1-0和CL1-5F4中差異性最大。
我們進一步將RGS11基因轉植入pGEX載體中,在大腸桿菌(JM-109)中表現此融合蛋白(GST-RGS11),經由表面有Anti-GST的beads純化,使用此純化過的蛋白做為抗原,注射到兔子的體內產生多株抗體。經由西方墨點法(Western blot)的初步分析,我們發現兔子所產生的多株抗體可以辨認GST-RGS11以及RGS11,不過無法證實CL1-0和CL1-5F4細胞有RGS11蛋白的存在。這其中的原因尚不明瞭,不過此抗體在未來可以用來進行CL1-0和CL1-5F4兩個細胞株的功能性測試,即可更加確認RGS11與肺腺癌轉移之間的關係。
