Abstract
由於無法有效抗菌、熱休克蛋白 (HSP) 表達受阻、血管生成受限以及持續的免疫調節和炎症加劇,糖尿病傷口難以恢復。光熱響應還原氧化石墨烯 (rGO) 具有抗菌和 HSP 促進活性。奈米級血小板衍生的細胞外囊泡 (NPV) 具有豐富的生長因子和細胞因子,可促進細胞增殖、血管生成、免疫和炎症調節。然而,單一藥物治療在其遞送效率和功效方面受到限制。聚合物生物工程可以通過結合精心挑選的材料和藥物來創造協同或互補的生物工程治療解決方案,從而提高單一藥物療法的有限有效性。在此,rGO、明膠 (Gel) 和藻酸鹽 (Alg) 被用來製造具有令人滿意的機械性能、多孔結構、生物降解性和溶脹比的 rGO 配位水凝膠 (GelAlg@rGO)。 GelAlg@rGO 被用作糖尿病傷口敷料,用於持續釋放 NPV,並與複合 GelAlg@rGO 水凝膠結合形成 GelAlg@rGO-NPV。 GelAlg@rGO-NPV 具有生物化學和生物物理學特徵。在 STZ 誘導的糖尿病大鼠傷口模型中,GelAlg@rGO-NPV 水凝膠有效降低炎症標誌物表達,調節免疫反應,促進血管生成,從而促進糖尿病傷口癒合。有趣的是,GelAlg@rGO-NPV 水凝膠增加了與細胞保護通路相關的 HSP 的表達。工程化的 GelAlg@rGO-NPV 水凝膠具有作為傷口敷料的潛力,能夠調節糖尿病傷口的免疫反應、炎症、血管生成和毛囊再生,最終導致慢性傷口更快癒合。
