Abstract
甲基乙二醛(Methylglyoxal, MGO)為目前已知與損傷具有正相關的物質,可藉由葡萄糖、胺基酸或是脂肪酸代謝合成。脂肪酸代謝的過程會產生D-三羥基丁酸(D-3HB),然而受限於其鏡像異構物L-三羥基丁酸(L-3HB)含量極低及分析上的限制,目前尚無相關文獻探討MGO與3HB之鏡像異構物的關聯性。本實驗採用馬兜鈴酸(AA)誘導小鼠腎損傷,研究不同劑量的AA造成的腎損傷對MGO與D-及L-3HB的濃度的影響。將雌性C3H / He小鼠隨機分為4組,除Normal組外,其餘3組(AAL、AAM、AAH)分別飲用低濃度AAI(4 μg/ ml)、中濃度AAI(8 μg/ ml)及高濃度AAI(12 μg/ ml)。實驗為期56天,動物犧牲後,取其血清及腎臟均質液加入螢光衍生化試劑 DDP或NBD-PZ進行螢光衍生化反應,再利用螢光高效能液相層析儀分析檢品中MGO與D,L-3HB的含量。實驗結果顯示,與Normal組相比投與不同劑量AA(AAL、AAM及AAH),其腎組織及血清中MGO含量均有增加趨勢(腎MGO:23.58 ± 5.85 vs. 25.39 ± 9.33 vs. 49.11 ± 9.99 vs. 60.40 ± 10.95 μg/g protein, p < 0.05;血清MGO:309.2 ± 136.5 vs. 1047.8 ± 249.7 vs. 941.4 ± 349.0 vs. 1418.1 ± 645.8 μg/L, p < 0.05),與組織損傷量化程度相符合。然而,(D,L)-3HB在腎組織中的濃度,不因投與不同劑量AA而有差異,推測乃因AA間接抑制腎臟中脂質的代謝所致。血清中L-3HB的濃度,與Normal組相比AAL及AAM組顯著增加,而在AAH組卻沒有統計上的差異(5.71 ± 1.35 vs. 16.76 ± 4.51 vs. 19.92 ± 9.24 vs. 12.76 ± 5.82 μM, p < 0.05),推測因損傷較為嚴重,需藉由TCA cycle代謝產生更多ATP提供損傷修復。因此我們認為同時監測血清MGO、L-3HB及creatinine,可幫助了解腎損傷的嚴重程度。當輕度及中度腎損傷時,血清L-3HB及MGO會顯著的增加;當重度腎損傷時,則發現血清MGO及creatinine會顯著增加。
