Abstract
本實驗室先前研究發現,溴化吳茱萸次鹼衍生物具有抗發炎作用,而目前已知的抗發炎藥物具有一個共通性,可抑制癌細胞的上皮細胞的上皮-間質轉化(Epithelial- Mesenchymal Transition, EMT)。在發炎的微環境中,TGF-β1和HIF-1會調控EMT的指標因子TWIST和SIP1的表現。EMT的主要現象包含癌細胞的侵入與遷移,先前研究發現溴化吳茱萸次鹼衍生物可以抑制Raw264.7小鼠巨噬細胞因為LPS刺激所引起的發炎現象,因此我們研究了溴化吳茱萸次鹼衍生物在抗EMT與抗癌轉移的活性。實驗結果發現,溴化吳茱萸次鹼衍生物可以降低在Raw264.7細胞的發炎指標蛋白iNOS與COX-2的表現,也能降低A2780卵巢癌細胞裡的EMT指標蛋白Vimentin的表現,也發現溴化吳茱萸次鹼衍生物能抑制A2780癌細胞的侵入與遷移活性,根據以上結果,溴化吳茱萸次鹼衍生物對於發炎所引起的EMT與癌轉移具有抑制作用,可成為一個新的治療策略。實驗室先前研究發現,溴化吳茱萸次鹼衍生物具有抗發炎作用,而目前已知的抗發炎藥物具有一個共通性,可抑制癌細胞的EMT作用,因此測試了溴化吳茱萸次鹼衍生物是否同樣具有抑制EMT之能力、對EMT指標蛋白的影響。
