Abstract
本研究主要目的是在探討乳癌與Annexin A6的關聯性。過去的臨床文獻指出透過利用數種不同篩選技術找尋與癌症相關的基因,發現Aneexin A6跟乳癌是具有相關性的,但文獻僅專注在篩選可作為診斷用的標誌,卻沒有深入探討Annexin A6在乳癌中扮演著如何的角色。
本研究在臨床上,利用乳癌病人的檢體,經由Real-Time PCR
定量mRNA的表現量,發現Annexin A6在正常組織表現量比在乳癌腫瘤組織中多,大約佔58%。更進一步利用雷射顯微擷取(LCM)和免疫組織化學染色(IHC)的方法,確認其再現性及表現分佈。另一方面,
利用實驗室現有的乳癌細胞株,探討其調控作用機制。近年來有研究指出, Annexin A6可能扮演一個抑癌因子的角色,為了探討這樣的機制是否也會發生在乳癌上面,透過細胞存活率測定實驗,將Annexin A6 si-RNA送入乳癌細胞當中,觀察其乳癌細胞生長的情形,發現Annexin A6具有抑制乳癌細胞生長的能力。接著當我們加入表皮生長因子EGF到乳癌細胞之後,發現Annexin A6的表現量上升,並且利用質膜分離的實驗,觀察到Annexin A6會移位到細胞膜上面,因此使用免疫沉澱法(IP),發現Annexin A6會與細胞膜上的EGF接受器反應。因此,推論Annexin A6在乳癌生長中可能扮演很重要的角色。
