Abstract
  多甲氧基類黃酮(polymethoxyflavones),廣泛存在於柑橘類水果(citrus fruits)的果皮中,如檸檬、橘子等其他柑橘類水果。Nobiletin有許多生理功能,包括抑制腫瘤形成(anti-carcinogenic)、抗發炎(anti-inflammatory),神經保護(neuroprotection)、抗粥狀動脈硬化(anti-atherogenic)及預防過度凝血(anti-thrombotic)的發生,但nobiletin對血小板的作用機制尚未被進一步的研究。因此,我們首次想要研究nobiletin是否會抑制人類血小板的活化。
  本研究發現,以collagen (1 μg/ml) 及arachidonic acid (60 μM) 當作刺激劑時,nobiletin可濃度相關性的抑制血小板的凝集。Nobiletin (15和30 μM) 也會抑制在collagen 刺激下,血小板內的ATP釋出及鈣離子移動。此外 , nobiletin也對於collagen 刺激下所引起的PLCγ2、p47 protein、AKT 及mitogen-activated protein kinases (MAPKs)的磷酸化也有抑制作用。有趣的是,我們也發現nobiletin會引起vasodilator-stimulated phosphoprotein (VASP)的磷酸化,但在adenylyl cyclase inhibitor SQ22536 及 guanylyl cyclase inhibitor ODQ處理下卻不會使血小板的凝集現象被回復。表示nobiletin 導致VASP的磷酸化是一種新的機制。
  最後,由我們的實驗結果首次發現,nobiletin的確可以抑制血小板的凝集。代表nobiletin也許能提供一個治療的潛力去預防或治療心血管疾病。
