Abstract
免疫系統為身體遭遇外來抗原所產生的一種防衛機制,主要分成先天性免疫系統 (innate immune system) 以及後天性免疫系統 (adaptive immune system),各有其相當複雜的防衛機轉,若是免疫系統異常,就會產生疾病。一旦免疫系統功能過高時,便會使得免疫細胞辨識到自體細胞並攻擊他們。此現象常發生於器官移植或自體免疫系統異常,常見的自體免疫系統異常疾病,如類風溼性關節炎、紅斑性狼瘡等。
檜木醇 (hinokitiol, ??-thujaplicin),俗稱為芬多精,存於柏科類植物心材之酸性結晶酚酮化合物,目前研究已發現其抗菌活性強及抗發炎之作用,並能抵抗多種外來病原,但對於免疫系統方面的研究仍然不明確。因此,本篇研究著重於檜木醇調控脾臟淋巴球細胞活化之作用。
我們研究結果證實檜木醇 (1-5?n?嵱) 會抑制由Con A (10 μg/ml) 所誘導的脾臟淋巴球之增殖反應,檜木醇主要是藉著增加 p21 蛋白的表現量及影響細胞週期的調控蛋白 (如Cyclin D、Cdk4及E2F1)使得淋巴球細胞生長停滯在G0/G1 phase,而不是藉由影響粒線體膜電位來使得淋巴球細胞凋亡。本篇研究結果顯示檜木醇確實會抑制淋巴球細胞增生,表示檜木醇可能在未來可做為調控免疫功能之藥物,對於器官移植及自體免疫疾病患者的治療與研究有很大的幫助。
