Abstract
此研究第一部分為評估飛梭鉺雅鉻雷射與飛梭二氧化碳雷射對經皮傳導小型干擾RNA (siRNA) 與質體DNA (plasmid DNApDNA) 載體的影響。體外穿透實驗以直立式擴散裝置(vertical diffusion cell) 分析藥物濃度。活體實驗以螢光顯微鏡 (fluorescence microscopy) 和雷射掃描式共軛焦顯微鏡 (confocal laser scanning microscopy, CLSM) 來觀察藥物滲透及分布情形。藉由飛梭雷射使皮膚表面剝離可使Fluorescein isothiocyanate – dextran (FD) 和siRNA經由皮膚吸收的程度明顯增加。在以飛梭鉺雅鉻雷射,能量5 J/cm2處理組之 FD-10 (10 kDa)和 siRNA的體外經皮穿透速率是未經處理組56倍和11倍。而以飛梭二氧化碳雷射,能量4 mJ,微點密度400 spots/cm2處理組之FITC-dextran和siRNA的經皮吸收量高於未經處理組42倍和12倍。從螢光顯微鏡和雷射掃描式共軛焦顯微鏡的影像呈現,可觀察到siRNA和pDNA經飛梭二氧化碳雷射處理後經皮吸收的螢光強度與藥物滲透深度有增加的現象。根據螢光顯微鏡和毛囊滲透量測定,可發現毛囊為皮膚經飛梭雷射處理後藥物穿透的一個重要途徑。另外也觀察到經飛梭鉺雅鉻雷射模式處理的siRNA毛囊蓄積量比經飛梭雷射模式處理組來的明顯。雷射也降低了E-cadherin在表皮中的表現,顯示在siRNA經皮吸收時的角質和表皮障蔽可藉由雷射處理而減少。第二部分再評估飛梭式電波後對經皮傳導小型干擾RNA (siRNA)與大分子胜肽 (peptide)的影響,並探討了皮膚結構與大分子趁透物經皮穿透間的相關機制。施予雙極電波後的經皮水分散失量為正常皮膚的3倍。經電波處理後投予大分子胜肽的經皮穿透累積量與穿透速率分別高於未施予電波的控制組3與23倍。siRNA10和siRNA15的經皮穿透累積量也在施予電波處理後高出控制組6.2和2.6倍。在雷射掃描式共軛焦顯微鏡的影像顯示,電波可使大分子胜肽的經皮穿透深度到達至少皮下74 μm深,也可讓siRNA的經皮穿透深度到達約皮下50 μm深。此研究第一次顯示出藉由飛梭雷射裝置的幫助可顯著的增加siRNA與pDNA的經皮穿透量與經皮穿透深度。也證實了經由電波可協助大分子胜肽與siRNA經皮傳遞。
