Abstract
本次實驗以高脂的致肥胖飲食誘發大鼠肥胖,並觀察同時給予Antroquinonol是否可以達到預防肥胖和腹部脂肪堆積的作用,並探討可能影響的能量代謝機制。實驗分為四組:控制組、肥胖飲食組、低劑量Antroquinonol介入組 (25 mg/kg BW)及高劑量Antroquinonol介入組 (50 mg/kg BW)進行為期12週的實驗。結果發現肥胖飲食組體重與腹部脂肪重量皆顯著高於控制組,而在Antroquininol介入後則顯著降低,此外在肥胖飲食組可觀察到由腹部脂肪堆積所導致的代謝異常現象包含有:adipokines的分泌異常、肝臟脂質異位堆積、肝功能異常和肝臟分泌的促發炎激素的濃度顯著上升,然而以上現象在Antroquinonol介入後均有顯著改善的作用。進一步探討其可能機制發現Antroquinonol可透過抑制肝臟中fatty acid synthase的mRNA表現量而減少肝臟中脂質的生成作用,另外Antroquinonol也具有增加肌肉中FNDC5 (Fibronectin type III domain-containing 5)蛋白質表現量的作用,可能具有增加體內生熱作用的功能。因此,由本次的實驗結果發現Antroquinonol可藉由抑制肝臟中脂質生合成和增加體內生熱效應,達到改善經由致肥胖飲食所誘導肥胖大鼠腹部脂肪堆積和代謝異常的情形。
