Abstract
TI-11是一種新化學成分,從中藥天麻萃取而來,具有可以減緩神經退化性疾病的病程之能力。由於神經退化性疾病患者的運動及認知障礙,會造成臨床上常用之錠劑在用藥上較為不便,因此,設計一種經非口服途徑且長效的劑型是必須的。微球包覆技術自發展以來,一直是最重要的藥物制放系統之一,且已經應用在製藥工業上。本研究主要目的是利用PLGA包覆T1-11而成微球,藉由微球包覆技術,使T1-11能達到長效釋放。本研究利用不同性質之PLGA以w/o/w方式進行包覆,藉由改變製備參數,評估各項條件對微球特性之影響。在電子顯微鏡下,T1-11微球呈圓球狀,但表面會有孔洞結構。全部配方之微球的平均粒徑皆在5~45 μm之間,且隨著PVA濃度增加,粒徑有減少之趨勢。T1-11微球的乘載率會隨著PLGA濃度提高而上升,最高乘載率為25.1±3.0%。體外釋放試驗結果顯示T1-11自微球中的釋放可分為兩個階段:初期的快速釋放,以及在48小時後的緩慢釋放。體外釋放試驗結果顯示,T1-11自微球至14天的釋放比例約為50%,顯示利用微球包覆技術,可使T1-11達到長效釋放的效果。
