Abstract
腦神經損傷(brain injury or CNS injury)包括了創傷性腦外傷(traumatic brain injury, TBI)、腦中風(stroke or CVA)、腦部腫瘤(brain tumor)等不同致病原因,不論致病原因為何,都可以造成癲癇及自主神經系統功能降低的後遺症,創傷性腦外傷是一個高度複雜的多因子疾病,透過類大腦損傷的動物實驗能有利於了解關鍵的損傷機制。而現今治療泛發性及局部性癲癇的藥物-valproic acid(VPA)用於治療腦神經損傷,其為組織蛋白去乙醯抑制劑(histone deacetylace inhibitor),可調節染色體中組織蛋白及DNA並調控基因表現。本研究目的在於透過類大腦損傷的動物實驗評估valproic acid對於治療創傷性腦外傷之效力。本實驗為Controlled cortical impact (CCI) 腦外傷模型,實驗動物為大白鼠SD品系,利用固定撞擊條件速度4 m/s、時間500 msec及2.0 mm之破壞硬腦膜,並配合每天給予30mg/kg 劑量之valproic acid治療下,觀察腦外傷前後大鼠短期與長期時間下慣用前肢運動功能的變化。藉此實驗探討外給一個valproic acid是否能保護腦組織免於創傷後所帶來的腦部傷害以及對運動功能恢復的影響。本實驗結果發現,使用Valproic acid治療之動物,於腦外傷七天後的腦組織病理切片細胞損傷的情形有明顯的減少,並能趨緩發炎反應的產生;而在慣用前肢運動功能的表現上,valproic acid治療之動物可以改善腦外傷後神經行為的能力缺失,且能恢復肢體的協調性,對長期的運動功能有改善之效力。以上結果顯示Valproic Acid(30 mg/kg),可以減少腦組織免於創傷所導致的神經傷害,具有神經保護的功能。
