Abstract
14-去氧穿心蓮內酯(14-deoxyandrographolide)為爵床科(Acanthaceae)草本植物穿心蓮(Andrographis paniculata Nees)之一種活性成分,屬於ent-labdane結構的二萜類。穿心蓮是一種在亞洲地區廣泛作為傳統治療的中草藥,用於治療感冒、肝臟相關疾病、腸胃道感染及上呼吸道感染。近年文獻指出14-去氧穿心蓮內酯具有多種活性,包括:治療肝疾病、降血壓、抗凝血及舒緩子宮收縮等。文獻上對於穿心蓮內酯之合成研究已有許多報導,然而對於14-去氧穿心蓮內酯進行半合成研究之報導較少,促使我們進行14-去氧穿心蓮內酯的構造修飾。由於14-去氧穿心蓮內酯於穿心蓮的含量較低,因此本研究先利用穿心蓮內酯進行化學合成14-去氧穿心蓮內酯,接著以此為原料,進行化學半合成反應,得到不同取代基的3??-O-substituted、19-O-substituted及3??,19-O-disubstituted的ent-labdane類似物,合成化合物的結構皆經由核磁共振及質譜決定。於這些合成的化合物中,化合物6a對於肝癌細胞株SK-Hep1、HuH7及HepG2具有毒殺活性,且當化合物6a與sorafenib併用時,對肝癌細胞株SK-Hep1具有增加細胞毒性作用。此外,化合物6a併用sorafenib對於肝癌細胞株HuH7及HepG2的細胞毒殺活性試驗正在進行中。
