Abstract
多形性膠質母細胞瘤 (GBM) 是最具侵襲性的腦腫瘤。快速遷移是
GBM 細胞的典型特徵。緩激肽受體 B1 (BDKRB1) 是緩激肽驅動的鈣
離子通道。我們之前的研究表明,緩激肽-BDKRB1 軸通過激活
MAPK-NFκB-水通道蛋白 4 通路參與 GBM 細胞的遷移。在這項研
究中,我們進一步旨在評估 BDKRB1 的拮抗劑 R715 對 GBM 細胞
遷移的功效及其可能的機制。 GBM 細胞暴露於 R715 不影響細胞
形態和細胞活力。用緩激肽處理 GBM 細胞可刺激細胞遷移。相反,
用 R715 預處理不影響細胞運動,但會顯著抑制緩激肽誘導的 GBM
細胞遷移。至於機制,緩激肽誘導的 GBM 細胞中細胞質 ROCK 蛋
白的增加被 R715 連續抑制。因此,本研究表明,BDKRB1 的下調可
以通過ROCK 通路抑制緩激肽誘導的 GBM 細胞遷移。
