Abstract
止血棉複合材於吸取大量血液同時,利用血液中所含有之生長因子以促進組織修復,亦提供患部之良好三維支架以利周圍組織再生。本研究以膠原蛋白/甲基纖維素/幾丁聚醣作為基材,採用冷凍乾燥法製備一新型止血棉複合材。其中膠原蛋白為人體細胞外基質中之主要成分,具有構成與修復組織、保持體內酸鹼平衡、水分平衡與電解質平衡等重要生理機能。探討止血棉複合材之生物相容性,遂根據ISO 10993測試標準進入產品之生物相容性評估並規劃兩大測試項目:體外試驗與動物性試驗。體外試驗包括:細胞毒性試驗、血液凝血性試驗,而動物性試驗包括:皮膚刺激試驗、皮膚過敏試驗、熱原試驗等。以此兩大主軸確認此止血棉複合材之生物相容性,期望達到具有細胞活性、止血情形良好之生醫材料。相關研究結果顯示;此止血棉複合材對MG-63與NIH3T3兩種細胞株不會產生立即性細胞毒性反應,血液凝血性試驗發現;血小板可易貼附於止血棉複合材表面且血液與止血棉複合材表面接觸20分後開始有凝血現象,可促進血液之凝結,於動物性試驗結果顯示,止血棉複合材不會造成皮膚過敏與刺激反應,以及未含熱原因子。
