Abstract
間質幹細胞已被證實會受到發展中的腫瘤吸引調控而移動至腫瘤處,並藉由分化成內皮細胞、基質細胞和纖維母細胞而幫助腫瘤血管新生和基質發展,所以使其極具潛力成為應用於癌症治療中的傳遞載體,因此本研究的第一部分就是應用間質幹細胞的趨癌特性結合蛋白質疫苗開發一種新的癌症治療平臺。我們使用針對第16型人類乳突病毒的E7蛋白所開發的蛋白質疫苗(PE(ΔIII)-E7-KDEL3)以及表現有E7抗原的間質幹細胞合併治療不具E7表現的腫瘤。結果顯示此合併療法可有效的抑制皮下及肺部轉移腫瘤的生長且會造成腫瘤細胞凋亡,我們也發現此治療效果和蛋白質疫苗誘發的專一性免疫效力成正相關,此部分研究證明了間質幹細胞可應用於追蹤腫瘤並媒介PE(ΔIII)-E7-KDEL3蛋白質疫苗的治療效力,我們認為這樣的策略可成為開發癌症的廣效型治療應用的平台。然而於第一部分的研究中我們另外發現了間質幹細胞可能會與癌細胞產生融合的現象,而間質幹細胞除了前述的幫助腫瘤基質發展的特性外,也被進一步證實會直接和癌細胞作用而促進腫瘤進程。間質幹細胞和癌細胞都被發現具有高度的細胞融合特性,且幹細胞被證實會透過細胞融合來進行組織修復,細胞融合是一種受到高度調控且非常複雜的細胞行為。然而間質幹細胞是否會透過細胞融合和癌細胞產生交互作用進而影響癌症的發展與進程尚不明瞭。所以本研究的第二部分就是想探討幹細胞融合對於癌症的作用影響,我們將肺癌細胞和間質幹細胞進行直接培養,接著利用兩種抗生素篩選、流式細胞儀分選雙螢光以及單細胞選殖方式挑選出融合細胞。結果顯示肺癌細胞會和間質幹細胞產生自發性的細胞融合且肺癌細胞會因為間質幹細胞融合所產生的基因重編成作用而具有幹細胞特性,我們進一步的發現FOXF1基因會調控基因重編成過程中所改變的細胞週期和上皮-間質轉化現象。總括來說間質幹細胞融合透過腫瘤基質效應參與癌症發展進程而非直接增強癌細胞的惡性表徵,此結果也提供了一個關於間質幹細胞可能用其它機制途徑影響癌症發展進程的新概念。
