Abstract
蛋白質二硫鍵異構酶在血栓形成與膠質母細胞瘤中扮演重要的的角色。此外,第二類組蛋白去乙醯酶與神經退化性疾病的病理機轉有關。本論文著重於這些酶的強效抑制劑之發現與優化,並探討其治療潛力及結構-活性關係
I. 開發新型蛋白質雙硫鍵異構酶抑制劑
PDI 透過催化半胱氨酸殘基的氧化還原反應來調控蛋白質穩定性,進而影響血栓與膠質母細胞瘤等病理過程。儘管已有一些小分子顯示出良好的 PDI 抑制活性,然而進入臨床試驗的候選化合物仍然有限。在第一章節中,將先前發現的 PDI 抑制劑 CPD 透過將其丙烯醯胺基團替換為不同的電子親和基團進行優化,成功開發了基於 2-三氟甲基丙烯酰胺的化合物。其中,化合物 14d 顯示出強效的 PDI 抑制活性 (IC50 = 0.48 μM),並具有顯著的抗血栓活性。第二章節中,為了改善抗膠質母細胞瘤活性,化合物 16d 進一步優化,並開發出帶有雙 2-氟甲基丙烯酰胺基團的衍生物。在這些衍生物中,化合物 40 顯示出強效的 PDI 抑制活性 (IC50 = 0.39 μM),並且相比 16d 具有更好的抗膠質母細胞瘤活性。第三章節中,對基於硝基苯乙烯的 PDI 抑制劑 HPW-RX40 進行結構修飾,得到了化合物 45a,其對 PDIA1 的選擇性明顯提高。進一步的結構修飾得到化合物 45v,該化合物顯示出顯著的抗 PDI 活性 (IC50 = 0.35 μM)。此外,我們將 α, β-不飽和硝基基團替換為可逆的邁克爾受體,得到了化合物 49,該化合物展現出卓越的 PDI 抑制活性 (IC50 = 0.006 μM)。
II. 開發強效的第二類組蛋白去乙醯酶抑制劑
第二類組蛋白去乙醯酶是阿茲海默症的潛在治療標的物。在此研究中,基於先前的研究結果,將吩噻嗪衍生物替換為具有 C-3 和 C-4 位點取代基的吩噁嗪化合物。C-4 位點取代衍生物表現出強效的第二類 HDAC 抑制活性,其中化合物 56d 展現了最高的抑制能力(IC50 = 3–870 nM)。此外,56d 在亞微摩爾濃度下有效保護神經細胞免受 H2O2 誘導的毒性,且未顯現顯著的細胞毒性,顯示其作為抗神經退化性藥物的潛力。

這項綜合性研究為設計選擇性與強效的 PDI 及第二類 HDAC 抑制劑提供了理論基礎,展現了其在血栓形成、膠質母細胞瘤及神經退化性疾病治療中的潛在應用。
