Abstract
發炎反應(inflammation)是人體對於外界刺激或是組織傷害所做出的良性生理反應,最後使感染被控制並且使組織回復正常的結構及功能。然而,長久而持續的發炎反應是導致很多疾病產生的原因,例如支氣管炎、腸炎、胃炎、多發性硬化症、類風濕關節炎等。因此,減緩身體過度發炎反應所造成的身體損害變成為了新藥開發的準則之一。冬蟲夏草(Cordyceps sinensis)在醫學領域的研究中被證實了有包括抗氧化、降血糖、抗癌症、調節免疫反應以及抗發炎的作用;CME-1是一新穎冬蟲夏草菌絲體多醣體萃取物,首度被分離與鑑定,主要以半乳糖及甘露醣組成,分子量為27.6 kDa的低分子量多醣萃取物。在先前的研究中我們發現,在巨噬細胞中CME-1可濃度相關性的抑制LPS (Lipopolysaccharide)所誘發的iNOS表現,並且其機轉可能是透過抑制IκB-α的降解及p65、Akt、Erk、JNK、p38之磷酸化而來,除此之外,我們也發現先給予PP2A抑制劑okadaic acid會明顯降低CME-1對發炎因子iNOS的抑制作用,本篇論文將對CME-1與PP2A之間的關係做進一步的探討。事先給予中性鞘磷脂酶抑制劑3-O-Methyl-Sphingomyelin (3-OME)會明顯降低CME-1對發炎因子iNOS的抑制作用、IκB-α的降解及p65、Akt、Erk、JNK、之磷酸化,CME-1也會提升PP2A的甲基化,結果顯示CME-1可以活化PP2A,可能藉由神經醯胺調控PP2A來抑制過度的發炎反應,也暗示CME-1是一個對於治療發炎相關疾病上具有潛力的藥物。
