Abstract
子宮肌瘤 (Leiomyoma)為婦女常見的疾病,在台灣,約有四分之一的女性患有肌瘤,臨床症狀為陰道出血、流產及不孕現象;目前認為肌瘤與雌激素調節失衡、細胞外基質過度增生及幹細胞調控有關。褪黑激素 (Melatonin)為大腦內松果體所產生的內生性荷爾蒙,可調節生理時鐘及雌激素分泌;文獻指出,Melatonin具有抗氧化、抗發炎、抑制癌細胞生長或轉移等作用,但對於子宮肌瘤抑制方面,文獻尚未探討。因此本研究目的為:探討Melatonin是否對子宮肌瘤細胞生長及細胞外基質堆積具有影響。本研究介入Melatonin於子宮肌瘤 (ELT-3)及平滑肌細胞株 (UtSMC),並利用側群細胞及幹細胞標的物所分選之肌瘤幹細胞進行實驗,藉由MTT assay、細胞計數、活細胞染色和細胞形態檢測增生;再利用流式細胞儀檢測其細胞週期及細胞凋亡,而相關蛋白質表現量以西方墨點法及細胞免疫螢光染色分析。由研究結果可知:1.Melatonin抑制ELT-3增生、細胞形態及肌瘤幹細胞的分選比例;2.介入後使肌瘤細胞週期增加sub G1期比例、及增加肌瘤細胞早期和晚期凋亡的分布;3.最終使細胞外基質相關蛋白表現量減少。Melatonin藉由細胞週期sub G1、細胞凋亡增加及細胞外基質相關蛋白質表現量減少,來抑制子宮肌瘤增生。
