Abstract
腫瘤微環境,是指包圍腫瘤的系統性組織。腫瘤細胞與其周圍組織之間的交互作用,是決定癌症進展的重要因素。在癌症進展的過程中,腫瘤微環境中的免疫系統扮演著關鍵角色。腫瘤細胞能夠形成具有免疫抑制特性的微環境,從而抑制免疫系統對腫瘤細胞的辨識與清除功能。因此,了解具有免疫抑制特性的腫瘤微環境中的作用機制,對於優化免疫療法在癌症治療中的應用是至關重要。本研究聚焦在介白素-1受體抗頡質 (IL-1Ra,基因名稱: IL1RN),其透過抑制介白素-1 (IL-1)細胞因子誘導的介白素-1受體 (IL1R1)活化,進而抑制下游的訊息傳遞調控機制。因此,IL-1Ra被認為具有抗發炎的功能,並已在臨床上應用於治療與發炎相關的疾病。儘管發炎已經被視為是癌症的一個重要標誌,IL-1Ra 在腫瘤生物學中的具體功能仍然不明確。一些研究表明,IL-1Ra可以透過抑制 IL-1β細胞因子誘導的發炎機制來抑制癌症進展; 然而,卻也有研究顯示,IL-1Ra 可能透過抑制抗腫瘤的免疫功能而促進腫瘤生長。為了解決這個問題,我們採用了兩種公認的免疫抑制型腫瘤模型:前列腺癌和胰腺導管腺癌(PDAC)。這兩種類型的腫瘤對現有的免疫檢查點抑制劑均呈現耐藥性,並被歸類為"冷"腫瘤。我們假設IL-1Ra的抗發炎功能會參與其中的機制,並且也想探討在腫瘤微環境中的細胞組成以及能表達IL-1Ra的細胞。為了完成這個研究,我們建立了小鼠腫瘤模型,其可以表現TME的特性,之後再利用各種體外實驗方式來驗證IL-1Ra的促進腫瘤功能。基於這些研究發現,我們證明在TME中的許多細胞 (包含單核細胞或是腫瘤細胞)利用IL-1Ra參與調控的功能促進了免疫抑制微環境的形成,因而促進了腫瘤的進展。綜合來說,我們的研究強調,IL-1Ra是一種新穎的免疫抑制機制調節因子,未來有可能成為"冷"腫瘤的預後標誌和治療標靶。
