Abstract
根據我們研究小組的近期發表的結果,二氫?啶(Dihydropyridine)與發炎因子IL-1β會促進睪固酮受體(androgen receptor: AR)活化,進而驅動睪固酮受體/connective tissue growth factor (CTGF)/collagen axis。根據文獻回顧,AR會導致轉錄因子slug活化,促進TGF-β,導致纖維化(fibrosis)現象,與臨床觀察到之現象符合。因此本實驗設計假設IL-1β及nifedipine會藉由睪固酮受體活化slug進而啟動增值細胞核抗原促使牙齦細胞增生導致纖維化。我們將闡明slug和PCNA在牙齦增生細胞中的分子機制,確立受IL-1β/或二氫?啶刺激之纖維母細胞中,對於睪固酮受體活化與slug的調節機制進而調控增值細胞核抗原的增加表現。並且建立起在牙齦增生纖維母細胞中,AR/slug/PCNA axis上下游關係。本實驗結果結果確立二氫?啶誘發增生之牙齦母細胞中,AR位於轉錄因子slug上游,並證實AR可調控slug之表現,進而調控增值細胞核抗原之生成。
