Abstract
阿茲海默症(Alzheimer’s disease,AD)為老年人中最常見的神經退化性疾病,其主要症狀為持續性的認知功能喪失、行為偏差及最終導致死亡。目前認為造成神經退化的主要原因與腦中過多的乙型類澱粉蛋白(β-Amyloid peptide,Aβ)有密切關係,Neprilysin(NEP)及Insulin-Degrading Enzyme(IDE)被視為腦中主要代謝Aβ之酵素,促進其活性可做為一具潛力之治療方式。本研究合成一螢光淬滅受質胜肽來偵測SH-SY5Y人類神經瘤母細胞上NEP和IDE活性,以提升酵素活性做為篩選的依據,對269株放線菌發酵液進行篩選,由此探討台灣本土放線菌液運用在AD上的可能性。結果顯示有4株放線菌液可提升酵素活性高達兩倍,以發酵穩定較好之菌液B79做進一步篩選,結果顯示濃度為5 μg/ml時,其EA萃取層所分離出來的第1、2和5管可提升酵素活性達1.5倍以上,當中更以第1管可提升至2.7倍,由此證實台灣本土放線菌運用在AD上的可行性。
