Abstract
近年研究發現使用臨床奈米藥物進行乳癌治療,常因腫瘤周邊纖維母細胞(tumor-associated fibroblast,TAF)提供其物理屏障,造成奈米藥物治療效果不彰。本實驗室過去已將可專一性標靶表現人類上皮因子接受體第二蛋白(human epidermal growth factor receptor 2,HER2)之乳癌細胞的乳癌專一性抗體(anti-HER2 scFv)和可標靶腫瘤周邊纖維母細胞表現之纖維母細胞活化蛋白(fibroblast activation protein,FAP)的抗纖維母細胞抗體(anti-FAP scFv),與人類化抗甲氧基聚乙二醇抗體(anti-mPEG Fab)結合,成功建立三功能抗體α-HER2/α-FAP/α-mPEG TsAb,能一步驟以非共價鍵方式結合上奈米藥物Lipo-Dox的mPEG,使Lipo-Dox能同時專一性標靶乳癌細胞和腫瘤周邊纖維母細胞。我們利用哺乳細胞量產系統(Expi293TM system)及鎳離子層析管柱(Nickel column)成功量產並純化這些抗體。以蛋白質電泳證實其蛋白質表現及摺疊皆正常;利用流式細胞儀證實三功能抗體α-HER2/α-FAP/α-mPEG可同時結合乳癌細胞及纖維母細胞;並利用ELISA證實三功能抗體可專一性結合上mPEG。重要的是,活體外細胞毒殺實驗證實,α-HER2/α-FAP/α-mPEG TsAb可提升奈米藥物(Lipo-Dox)毒殺單獨及以不同比例培養的HER2+乳癌細胞、FAP+纖維母細胞之能力。根據上述成功的結果,本研究將提供一個創新的乳癌治療策略,將三功能抗體α-HER2/α-FAP/α-mPEG TsAb與任何mPEG-奈米藥物一步驟混合反應,即可快速轉變為同時標靶HER2及FAP的奈米藥物,能有效標靶並毒殺HER2+乳癌細胞及FAP+腫瘤周邊纖維母細胞,大幅提升臨床乳癌治療之成效。
