Abstract
結締組織生長因子(CTGF)屬於CCN家族的一員,是一種分泌型蛋白,而它在許多生物功能上扮演了重要的角色,例如調控纖維化、細胞凋亡、黏附、移行、分化、增生、血管新生、軟骨生成等。已知CTGF可增加許多前炎性因子的產生,但未有許多研究針對CTGF誘使發炎性細胞激素做討論。在本實驗先前的研究已知,結締組織生長因子刺激人類CD14+單核球細胞分泌IL-8。在本篇研究中接續先前的實驗進一步探討結締組織生長因子CTGF誘使IL-8基因表現機制,在轉錄調控及後轉錄調控部份。我們以HEK293T細胞株做為實驗模式,轉染入野生株或突變之IL-8 promoter調控區控制之luciferase報告質體,給予CTGF刺激,觀察CTGF對於IL-8 promoter轉錄調控的機制。實驗結果發現IL-8 promoter上 AP-1及NF-κB binding site對於CTGF刺激IL-8轉錄是重要的,且CTGF會誘使JNK、ERK及p38 MAPK路徑的活化,進一步活化轉錄因子AP-1及NF-κB,誘使IL-8基因的轉錄。文獻指出,IL-8 mRNA 3’UTR區域具有許多AU-rich element,導致其mRNA較為不穩定。為了解給予CTGF刺激,是否會影響IL-8於後轉錄層次,此部分的實驗以293細胞株做為實驗模式,分別轉染入包含不同IL-8 3’UTR區域片段之luciferase報告質體,觀察在給予CTGF刺激下對於luciferase基因之mRNA穩定度的影響。實驗結果發現CTGF可能經由AURE independent途徑誘使IL-8 3’UTR mRNA變穩定,而我們也發現CTGF活化的MAPKs中,p38 MAPK路徑的活化,會參與在CTGF誘使IL-8基因後轉錄調控的機制中。
