Abstract
阿茲海默氏症 (Alzheimer's disease,簡稱AD),是一種常見的神經退化性疾病。老化是神經退化性疾病的危險因子,因全球走向高齡化社會,所以AD患者越來越多,隨著時間其病情進展甚至無法自理生活,使生活品質下降,造成社會的成本更加沉重。而目前的藥物無法根治此病,只能延緩其進展。因此如何預防以及治療是目前很重要的課題之一。目前被提出可以使用抗發炎療法。
故研究擬在小鼠星狀神經膠細胞中添加黃花蜜菜中的主要成分芹菜素 (Apigenin) 以及木犀草素 (Luteolin),探討其是否具有對抗脂多醣 (LPS) 及乙型類澱粉蛋白 (Aβ) 所誘導之發炎反應以及細胞保護的作用。
結果顯示,LPS濃度10-6M及Aβ濃度10-6M下對星狀神經膠細胞之毒性是可以確定的,並促使發炎因子TNF-α、IL-1β、COX-2和iNOS的mRNA表現量上升、 NO產量增加、並對星狀神經膠細胞造成氧化壓力。而以10-7M芹菜素以及10-7M木犀草素分別預先處理下,可以下調LPS以及Aβ刺激星狀神經膠細胞所誘導之發炎因子TNF-α、IL-1β、COX-2和iNOS的表現量下降將近1倍,並減少NO50%~70%的表現,所以減緩其氧化壓力,達到保護細胞的效果。
