Abstract
轉糖鏈球菌(Streptococcus mutans)是一種存在人類口腔中造成齲齒的主要致病菌之一,在口腔黏膜受損的情況下,細菌進入血液循環引發菌血症。當血液流經受損的心臟瓣膜時,S. mutans 會附著於瓣膜上,導致宿主誘發感染性心內膜炎(Infective endocarditis;IE),這是一種高致死率的疾病。S. mutans 的致病力與其生物膜形成能力密切相關,細菌在死亡過程中會調控自身溶解反應(autolysis)促使細菌釋放出細胞外 DNA(extracellular DNA; eDNA),為形成生物膜的所需的重要成份之一。在我們過去研究發現,細菌中的自體溶解素(AtlA)能夠調控感染性心內膜依賴 eDNA 所形成的生物膜,在 atlA 操縱組當中,我們發現 thmA 極性突變體表現出生物膜生成能力降低,並進一步發現 thmA 下游基因 Smu. 695 缺失會下調 AtlA 介導生物膜的形成。在本篇研究中發現,將 thmA、Smu. 694 以及 thmA-Smu. 694 分別轉化至 thmA 極性突變體(thmA polar),回補後的 thmA-Smu. 694 生物膜形成會比 thmA polar 少,有趣的是,當 thmA 回補到 thmA polar 則會導致細菌分離速度減緩形成更長的細菌型態,生物膜生成能力甚至比 thmA-Smu. 694 回補後更低,但是回補到野生株並沒有影響。過去結果顯示 Smu.695 與 thmA 間可能存在調控關係,然而當我們將 thmA 回補至 thmA-Smu. 695 突變體中,其細菌長度卻沒有發生改變。此外,我們進一步構建重組蛋白以探討 ThmA 的功能。結果顯示在不同濃度 IPTG 影響下,ThmA 在大腸桿菌中會以劑量依賴方式抑制其生長,說明 ThmA 對細菌存活具有負面影響。綜合上述結果,我們推測 ThmA 可能透過尚未明確的調控機制負向調節細菌生長與生物膜形成,並可能在 IE 的致病過程中扮演關鍵角色。
