Abstract
肥胖之人口在台灣與世界其他國家均逐年緩慢的增加,且男性之肥胖人口比例已遠超過鄰近之國家,與肥胖相關的心血管與代謝性疾病在十大死因中也佔有數個位子,對醫療資源與人民的生活都造成不小的影響。小葉葡萄是台灣特有種葡萄科植物,在過去的研究中顯示其具有抗菌、抗發炎、抗高血壓、抗凝血、抗肥胖、抗糖尿病與抗氧化等活性;在化學成分上,小葉葡萄富含二苯乙烯類化合物,文獻顯示在心血管保護、抗癌、抗肥胖與抗糖尿病上均有不錯的活性,其中 (+)-ε-viniferin為重要之二聚體,為形成其他寡聚體之重要單元,在過去研究中已顯示具有抗肥胖與干擾脂肪細胞分化之活性,但缺乏相關作用機制之探討。本實驗之目的為評估小葉葡萄熱水抽取物是否具有抗肥胖之活性與開發價值,並進一步對其根部所含 (+)-ε-viniferin抑制脂肪細胞分化之作用機制加以探討。結果顯示,在高油脂飲食誘導小鼠肥胖之模式下,每日經口投與小葉葡萄熱水抽取物(40 mg/kg,共計 36天)與其中所純化出來之 (+)-ε-viniferin(兩階段實驗,第一階段之劑量為 10 mg/kg,為第 1-38日;第二階段為 39-58日,劑量為 25 mg/kg),均能在不干擾小鼠進食能力之情況下,有效的抑制高油脂飲食所誘導之體重增加。而使用 (+)-ε-viniferin所進行的兩階段實驗,更進一步發現能有效的降低血糖、總膽固醇、低密度脂蛋白於血液中之濃度,以及降低腸系膜脂肪組織之身體體重佔比。且 (+)-ε-viniferin對於膽固醇生成之重要酵素 HMG-CoA還原酶具有明顯的抑制作用,其 IC50為 96 μM。另一方面,小葉葡萄熱水抽取物與 (+)-ε-viniferin對 3T3-L1前脂肪細胞經過八天分化於細胞內累積的脂質量均能有濃度依存性之抑制效果,進一步對 (+)-ε-viniferin研究發現,其可透過降低轉錄因子 C/EBPβ與細胞週期調控分子 cyclin A2之表現,達到對 3T3-L1細胞分化過程中之細胞擴增產生抑制,降低細胞分化與脂質之累積。本研究之內容顯示小葉葡萄熱水抽取物與其所含之 (+)-ε-viniferin在重量控制上具有開發之價值,值得後續之評估與研究。
