Abstract
憂鬱症是全球重要的心理健康議題,其致病機轉錯綜複雜,涉及多種生理與心理因素。腸腦軸線被視為研究情緒調控的重要方向,其中神經傳導物質、腸道屏障功能和腸道菌叢可能共同參與調節神經發炎反應,進而影響中樞神經功能與情緒行為。近年來,地中海型飲食因富含omega-3多元不飽和脂肪酸(PUFA)及omega-9單元不飽和脂肪酸(MUFA),被證實有助於改善情緒症狀的潛力。然而,其調控神經傳導物質代謝、神經發炎及腸道菌相影響情緒與行為的具體機制仍不明確。本研究旨在探討慢性非預期性壓力(CUMS)對大鼠腦部及腸道中色胺酸與酪胺酸相關代謝物之影響,並進一步評估介入omega-3 PUFA與omega-9 MUFA對腸腦軸線與憂鬱樣行為之潛在改善作用。實驗採用25隻Sprague–Dawley大鼠,隨機分為五組:控制組(N)、慢性非預期性壓力組(C)、藥物組(P)、魚油組(F)及橄欖油組(O)。實驗為期14週,魚油與橄欖油介入自實驗第1週開始,自第3週開始CUMS刺激,憂鬱症藥物Imipramine則於第9週介入。動物行為結果顯示,自第6週起除魚油組外,其餘組別在蔗糖偏好測試中均表現出明顯的失樂現象(p<0.05),第8週起此現象更趨明顯;強迫游水試驗亦顯示C組的不動時間顯著高於N組、P組及F組。標靶質譜分析結果指出,CUMS刺激下促使大腦皮質及腸黏膜中色胺酸代謝轉向kynurenine途徑。腸黏膜之tyrosine與dopamine濃度於所有受壓力組均顯著高於N組,且P、F、O組的dopamine顯著低於C組,同時伴隨大腦皮質中dopamine代謝產物HVA濃度上升。發炎指標則顯示,C組大腦皮質中GFAP與TNF-α皆上升,而F組與O組有降低的趨勢,同時大腦及腸道的IL-10皆呈現相反趨勢,反映攝取魚油與橄欖油具神經抗發炎潛力。腸黏膜蛋白分析中,魚油可改善腸道屏障功能損傷,並與橄欖油共同調節腸道菌相組成。3-hydroxykynurenine (3-HK)在腸道與大腦中的濃度呈正相關,顯示其可能在腸腦軸訊息傳遞中扮演關鍵角色。
