Abstract
Alpha-mangostin (MG) 為山竹中的主要成分之一,具有抗氧化、抗凋亡及調節血糖等作用。因此本研究欲探討在鏈脲佐菌素 (Streptozotocin, STZ) 誘導的胰島細胞功能障礙中,給予MG對於胰島細胞的保護作用與其粒線體功能及生合成之影響。以6 mM的STZ處理胰島細胞株 (RIN-m5F) 一小時後,將STZ於培養液中移除,並給予5-15 μM的MG培養二十四小時,再進行細胞存活率、細胞凋亡、細胞與粒線體內的氧化壓力、胰島素濃度、粒線體功能及其生合成之相關蛋白質與基因表現分析。發現STZ的介入會顯著地降低粒線體膜電位與減少細胞內胰島素含量,並增加細胞與粒線體內的氧化壓力,此外,亦會降低粒線體功能、生合成相關因子mitochondrial transcription factor A (Tfam) 的蛋白質表現與減少粒線體DNA拷貝數,造成胰島細胞凋亡與死亡,而給予5和10 μM的MG介入後能顯著地恢復粒線體膜電位、維持細胞內胰島素含量,且增加粒線體功能、Tfam蛋白質表現與粒線體DNA拷貝數,但無法降低其細胞與粒線體內的氧化壓力,卻不會因為氧化壓力的增加而造成細胞死亡。因此綜合上述顯示alpha-mangostin透過增加粒線體功能與其生合成,進而減少STZ所誘導之胰島細胞粒線體功能不全及細胞死亡的情況。
